1. NAD⁺ Precursor and Preiss–Handler Pathway
A 2024 multi-species study reported that trigonelline enters the NAD⁺ pool through the NAPRT-dependent Preiss–Handler pathway. Human serum trigonelline was lower in sarcopenia and correlated with muscle strength and oxidative-phosphorylation signatures, while causal muscle-function findings came from cells, C. elegans and mice rather than a human supplementation efficacy trial.
